The run will feature an eight-kilometre event and a four-kilometre race for the adult runners.
There will be a two-kilometre race for children aged 12 years or younger.
Athletics NSW and Port Stephens Council, as well as Raymond Terrace Athletics Club, are working together on the run.
"The Raymond Terrace Fun Run will become part of our newly-formed series of community fun runs being staged throughout NSW," Athletics NSW chief executive Greg Meagher said.
"The series is being labelled as Run NSW and invites runners of all abilities."
The run will start along the Hunter River adjacent to Vi Barnett Field and follow the river south along King, William and Hunter streets before turning left onto Swan Street and running out to a turnaround point.
"The route is fantastic," event organiser and Australian marathon representative Scott Westcott said.
Raymond Terrace Athletics Club is set to assist in the delivery of the run and it will add to a busy start to their formal athletics season.
A training program is available on the website and entries are now open.
