The 21-year-old woman was arrested by Manning-Great Lakes and Mid North Coast police at a North Haven motel on Monday, March 2, following inquiries into the death of 27-year-old man.
Emergency services were called to the intersection of Watt and Payton streets about 5.45am on Saturday after the man's body was found in the middle of the road with multiple stab wounds.
Port Stephens-Hunter Police officers established a crime scene, which was examined by specialist forensic police.
Following inquiries, the 21-year-old woman was arrested in North Haven for an outstanding warrant.
Police said it was believed that the man and the woman were known to each other.
Detectives from Port Stephens-Hunter Police District travelled to Port Macquarie Police Station on Monday and charged the woman with murder.
She was refused bail and is due to in Port Macquarie Local Court on Tuesday, March 3.
