The evening of acoustic and alternative tunes, market stalls, vintage clothing, art and good food will also be a chance to access information and help for mental illness.
The action will kick off at the Beach House Restaurant at One Mile Beach at 4pm on Saturday, May 3.
Event organiser Lisa Papasavvas said the venue was expected to fill up for yet another season.
"There's a great sense of community," she said.
Papasavvas said there would not be a time when there was no music playing.
Sixteen acts from across the Port will fill the main stage, an alternative stage inside and the acoustic stage outside from 4pm to 10.30pm.
Ranging from blues, to heavy metal, pop, modern alternative, mainstream and punk the night should appeal to all music palates, with an older blues band covering the Beatles and young acts performing their versions of songs often heard on Triple J.
Many of the bands will sell merchandise and albums on the night.
Gates open at 3pm for those wishing to shop at the market for knick-knacks or relax at an alternative therapy stall.
Visitors can purchase artwork by Port talent or shop for a bargain at the second hand and vintage stalls.
Intertwined with the festival-like event will be information stands on services that promote healthy and happy living, such as Tomaree Youth Community Action and a suicide prevention team.
Cancer Council donation boxes will also be posted around the venue.
Inside guests can choose the option of a set restaurant dinner which includes a starter, a main course and a drink for $25, or grab a $15 take-away pizza to sit outside and take in acoustic melodies. The kiosk will sell snacks.
Tickets cost $7 pre-sale or $10 at the door.

It is a lockout event so when patrons leave they will not be allowed back in.
