The trip is part of the Sister City program between Port Stephens and Tateyama, about two hours south of Tokyo.
Each year the Port sends runners to compete in the annual Wakashio Marathon, which is set to take place on January 25 next year.
Oonagh Treble, Tracey Spicer, Scott Brownlie and Clare van Doorn - all from the Nelson Bay Hash House Harriers - will compete in the event.
"We have heard that the course is pretty flat," Oonagh Treble said.
"We have been told from someone who has done the event that the hospitality is excellent."
Part of the experience includes attending meet-and-greets at local schools and a traditional tea ceremony.
Each person will be hosted by a local family.
"I have always wanted to see Japan," Scott Brownlie said.
"What better way to do so than to represent where I come from."
This hospitality will be reciprocated when residents from Tateyama come to visit Port Stephens.
The Sister Cities committee is also subsidising the runners' trip. It is offering $500 to each person to help with their expenses.
