The service, which is recited across 170 countries on the same day, will take place on Friday, March 6, from 10am.
The Rock's spokeswoman Sally Dover said the annual prayer day in Port Stephens would bring together people from all churches and denominations.
"We are expecting more than 100 parishioners to participate," Mrs Dover said.
"We welcome people from all churches in Port Stephens for a time of prayer and fellowship."
The worldwide host this year is the Bahamas with a focus on domestic violence.
"This is an issue affecting Port Stephens as much as the Bahamas or any other nation," Mrs Dover said.
"We are also asking attendees to dress up in the Bahamas' national attire, including colourful and elaborate scarves and headwear."
Last year's local host was the Uniting Church.
